Efektivitas Anti Mikroba Air Ion Perak terhadap Derajat Keasaman (pH) Streptococcus mutans

Abstract: Silver ion water has been known as an effective material for inhibiting and killing pathogenic bacteria. However, there are still few studies on silver ion water, especially those related to oral pH in dentistry. This study aimed to determine the effectiveness of silver ion water against Streptococcus mutans in decreasing the environmental pH. This was an experimental and laboratory study with a control group post-test only design, using Streptococcus mutans stereotype C strain UA 159 incubated in brain hearth infusion broth (BHIB). Samples were divided into eight groups with several concentrations of silver ion water (2.5 ppm, 5 ppm, 7.5 ppm,10 ppm, 12,5 ppm, 15 ppm) and two control groups (silver ion water 0 ppm and aquadest). All groups were incubated for 24 hours in 37oC, and then their pHs were measured with a pH meter. The normality test showed a p-value of >0.05, meaning that the data were normally distributed. The Mann-Whitney test showed a p-value of <0.05, meaning that there was a difference in each group. Significant increases of pH were found in silver ion water of 12.5 ppm (mean pH 5.670) dan 15 ppm (mean pH 6.772). In conclusion, silver ion water of 12.5 ppm and 15 ppm can effectively inhibit the decrease in pH caused by the metabolism of Streptococcus mutans.

Abstrak: Air ion perak telah dikenal efektif untuk menghambat dan membunuh bakteri patogen, namun penelitian tentang air ion perak masih sangat terbatas, terutama yang berhubungan dengan pH rongga mulut di bidang kedokteran gigi. Penelitian ini bertujuan untuk mengetahui efektivitas air ion perak menghambat Streptococcus mutans dalam menurunkan pH. Jenis penelitian ialah eksperimental laboratorik dengan group control post-test only design, menggunakan Streptococcus mutans stereotype C rantai UA 159 yang diinkubasi di dalam media brain hearth infusion broth (BHIB). Sampel dibagi dalam delapan kelompok, masing-masing diberi air ion perak sebanyak 2,5 ppm, 5 ppm, 7,5 ppm, 10 ppm, 12,5 ppm, 15 ppm, dan dua kelompok kontrol (air ion perak 0 ppm dan akuades). Seluruh kelompok perlakuan dan kontrol diinkubasi selama 24 jam dengan suhu 37oC, kemudian pH-nya diukur menggunakan pH meter. Uji normalitas menunjukkan p<0,05 yang berarti data berdistribusi normal. Uji Mann Whitney menunjukkan p<0,05 yang berarti terdapat perbedaan bermakna antar kelompok perlakuan. Efek peningkatan derajat keasaman (pH) yang bermakna terlihat pada pemberian air ion perak sebanyak 12,5 ppm (rerata pH 5,670) dan 15 ppm (rerata pH 6,772). Simpulan penelitian ini ialah air ion perak dengan konsentrasi 12,5 ppm dan 15 ppm efektif menghambat penurunan pH yang disebabkan oleh metabolism Streptococcus mutans.
