Pengaruh Pemberian Ekstrak Sirih Merah (Piper crocatum Ruiz & Pav) terhadap Pembentukan Zona Hambat Pertumbuhan Bakteri Streptococcus mutans

Abstract: Dental caries has the highest prevalence among all oral and health problems. Streptococcus mutans is the main bacteria that cause dental caries. Another alternative that can be used in preventing caries is traditional medicine, such as red betel leaves (Piper crocatum Ruiz & Pav). The efficacy of red betel is caused by the presence of some active compounds. This study aimed to analyze the effect of administering red betel extract on the formation of inhibitory zone for the growth of S. mutans. All samples of S. mutans were obtained from the microbiology laboratory of the Faculty of Medicine, Universitas Wijaya Kusuma, Surabaya. Sample size taken was at least four samples for each group using a simple random sampling technique. Data were analyzed using the analysis of variance test (ANOVA). The results showed that the diameter of inhibition zone was already visible at a concentration of 10%, with an average of 20.25 mm. Meanwhile, the highest inhibition zone diameter in the treatment group was observed in group P6 treated with red betel leaf extract at a concentration of 50%, with a diameter of 35 mm. In conclusion, the effective concentration of red betel leaf extract (Piper crocatum Ruiz & Pav) in inhibiting the growth of Streptococcus mutans bacteria is at a concentration of 50%.

Abstrak: Karies gigi memiliki prevalensi tertinggi dari seluruh masalah kesehatan gigi dan mulut. Steptoccocus mutans merupakan bakteri penyebab utama terjadinya karies gigi. Salah satu alternatif dalam mencegah terjadinya karies ialah penggunaan pengobatan tradisional, antara lain daun sirih merah (Piper Crocatum Ruiz & Pav). Khasiat sirih merah itu disebabkan oleh adanya kandungan sejumlah senyawa aktif. Penelitian ini bertujuan untuk menganalisis pengaruh pemberian ekstrak sirih merah terhadap pembentukan zona hambat pertumbuhan S. mutans. Bakteri S. mutans diperoleh dari Laboratorium Mikrobiologi Fakultas Kedokteran Universitas Wijaya Kusuma Surabaya. Besar sampel diambil minimal empat sampel untuk setiap kelompok dengan teknik simple random sampling. Analisis data penelitian menggunakan uji analisis varian (ANOVA). Hasil penelitian mendapatkan bahwa diameter zona hambat sudah terlihat pada pemberian ekstrak konsentrasi 10% dengan rerata diameter 20,25 mm, sedangkan diameter zona hambat tertinggi pada kelompok perlakuan P6 yaitu kelompok dengan pemberian ekstrak 50% dengan diameter 35 mm. Simpulan penelitian ini ialah konsentrasi efektif ekstrak daun sirih merah (Piper crocatum Ruiz & Pav) dalam menghambat pertumbuhan bakteri Streptococcus mutans pada konsentrasi 50%.
