Uji Efektivitas Larutan Prebiotik Buah Naga dalam Menghambat Pertumbuhan Porphyromonas Gingivalis secara in vitro

Abstract: Periodontal disease in pregnant women is related to an increased risk of pregnancy complications due to bacterial infections, particularly Porphyromonas gingivalis. Natural mouthwashes containing prebiotics are a promising alternative to support oral and periodontal health. This study aimed to assess the antibacterial efficacy of dragon fruit prebiotic solution against P. gingivalis. A disk diffusion method was employed using agar medium to observe the inhibition zone of P. gingivalis. Dragon fruit prebiotic solutions of 10%, 20%, 30%, and 40% were used as treatment groups; each was duplicated four times. The diameters of inhibitory zones were measured in millimeters, and the ANOVA test was used to examine group differences. The results showed that as the concentration increased, the inhibition zone diameter significantly increased. The mean diameters were 8.2 mm (10%), 12.5 mm (20%), 16.8 mm (30%), and 20.3 mm (40%). All groups showed significant differences with p-values less than 0.05. In conclusion, dragon fruit prebiotic solution demonstrates antibacterial potential against Porphyromonas gingivalis. The 40% concentration exhibits the highest effectiveness (very strong category), indicating its potential as an active natural mouthwash ingredient.

Abstrak: Penyakit periodontal pada ibu hamil dikaitkan dengan peningkatan risiko komplikasi kehamilan akibat infeksi bakteri, khususnya Porphyromonas gingivalis. Obat kumur berbahan alami yang mengandung prebiotik menjadi alternatif menjanjikan dalam mendukung kesehatan mulut dan jaringan periodontal. Penelitian ini bertujuan untuk mengetahui efektivitas daya hambat larutan prebiotik buah naga terhadap P. gingivalis dengan menggunakan pengujian in vitro. Metode difusi cakram pada media digunakan untuk mengidentifikasi hambatan pertumbuhan P. gingivalis. Larutan prebiotik buah naga diuji pada konsentrasi 10%, 20%, 30%, dan 40%. Diameter zona hambat diukur dalam milimeter (mm) dan dianalisis menggunakan uji ANOVA. Hasil penelitian menunjukkan bahwa seiring dengan peningkatan konsentrasi, diameter zona hambat meningkat secara bermakna. Rerata zona hambat ialah 8,2 mm (10%), 12,5 mm (20%), 16,8 mm (30%), dan 20,3 mm (40%). Analisis statistik menunjukkan perbedaan bermakna antar semua kelompok (p<0,05). Simpulan penelitian ini ialah larutan prebiotik buah naga memiliki potensi sebagai agen antibakteri alami terhadap Porphyromonas gingivalis. Konsentrasi 40% menunjukkan efektivitas tertinggi (kategori very strong), sehingga berpotensi dikembangkan sebagai bahan aktif dalam obat kumur alami.
