UJI EFEK ANALGESIA EKSTRAK DAUN KECUBUNG (Datura metel L.) PADA TIKUS WISTAR (Rattus norvegicus) JANTAN

Abstract: Jimson weed (Datura metel) is rich of various chemical compounds found in the roots, stems, leaves, fruits, flowers, and seeds. Jimson weed are used for asthma, dental pain, and anesthetic as well as analgesic medications. Analgesia is the loss of the sensation of pain without loss of other senses. Anesthesia is the reduction or removal of sensation for a while, so that surgery or other painful procedures can be performed. This study aimed to test the analgesic and anesthesia effects stage 2 and 3 of jimson weed leaf extract on male Wistar rats. This was an experimental study with a praexperiment-postes only (one shot case study) design. Jimson weed leaves were taken from Central Sulawesi. Its extract was made by using maceration method. Six male wistar rats were used as samples. The results showed that in response to the extract, 5 rats entered the first stage (analgesia) and only 3 rats entered the third stage. Conclusion: Jimson weed leaf extract had analgesic effects on male Wistar rats. The initial time and the length of time to be conscious were influenced by body weight, age, and time of adaptation.

Abstrak: Tanaman kecubung (Datura metel) kaya dengan berbagai senyawa kimia yang terdapat pada akar, tangkai, daun, buah, bunga, dan biji. Tanaman kecubung banyak dimanfaatkan antara lain sebagai obat asma, obat sakit gigi, obat bius, dan obat analgesia. Analgesia merupakan hilangnya rasa sakit tanpa kehilangan sensasi indera lainya. Anestesi merupakan pengurangan atau penghilangan sensasi untuk sementara, sehingga operasi atau prosedur lain yang menyakitkan dapat dilakukan. Penelitian ini bertujuan untuk menguji efek analgesia ekstrak daun kecubung pada tikus wistar jantan dan efek anestesi stadium 2 dan 3. Penelitian ini menggunakan metode eksperimental dengan rancangan penelitian praexperiment-postes only (one shot case study). Daun kecubung yang digunakan berasal dari Sulawesi Tengah yang dibuat ekstrak dengan metode maserasi. Penelitian ini menggunakan 6 hewan coba tikus wistar jantan. Hasil penelitian memperlihatkan bahwa kelima hewan coba menunjukkan respon terhadap esktrak daun kecubung dengan masuk pada stadium 1 yaitu stadium analgesia dan hanya tiga hewan coba yang sampai stadium 3. Simpulan: Ekstrak daun kecubung mempunyai efek analgesia pada tikus wistar jantan; waktu awal teranestesi dan lama waktu sadar dipengaruhi oleh berat badan, usia dan waktu adaptasi.
