PENGARUH KONSENTRASI ZAT PENGATUR TUMBUH THIDIAZURON (TDZ) TERHADAP PERTUMBUHAN PLANLET KENTANG INDUSTRI

This study aimed to determine the effect of Thidiazuron (TDZ) growth regulator concentrations on the in vitro growth of industrial potato plantlets from two cultivars, Ventury Agrihorti and Golden Agrihorti. The research was conducted at the Biotechnology Laboratory, Faculty of Agriculture, Sam Ratulangi University, Manado, using a factorial Completely Randomized Design (CRD) consisting of two factors: cultivar (V1 = Ventury, V2 = Golden) and TDZ concentration (0, 1, 2, 3, and 4 ppm), each with five replications. The observed variables included the number of leaves, number of shoots, plant height, and dry weight. The results showed that TDZ concentration significantly affected plantlet growth, particularly the number of shoots, which increased significantly in the Ventury Agrihorti cultivar at 1 ppm TDZ. The treatment without TDZ (control) produced the best results for plant height, number of leaves, and dry weight. The Ventury Agrihorti cultivar produced higher dry weight than Golden Agrihorti, while Golden Agrihorti exhibited better plant height. The negative effects of TDZ were presumed to be due to the decrease in medium pH after autoclaving, which affected hormone effectiveness and nutrient stability in the medium.
