Abstract: Fatigue represents a significant occupational health issue among co-assistants, which is influenced by work shift and high workload, potentially impacting performance and patient safety. This study aimed to analyze the influence of work shift and workload on fatigue among co-assistants at Prof. Dr. R. D. Kandou General Hospital, Manado. This was an observational analytical study with a cross-sectional design. Samples consisted of 108 co-assistants selected through purposive sampling. Shift was measured using self report questionnaire. Workload was measured using the NASA-Task Load Index (NASA-TLX) questionnaire, and fatigue was measured with the Swedish Occupational Fatigue Inventory (SOFI). Data were analyzed using the Spearman's test and ordinal logistic regression. The results showed that the majority of respondents had high workload (46.30%) and severe fatigue (60.18%). Bivariate analysis showed a significant positive relationship (p<0.05). Multivariate analysis showed a significant simultaneous effect of shift work and workload on fatigue (p<0.05), accounting for 38.2% of the variation in fatigue. The majority of respondents experienced severe fatigue and high workload. The ordinal logistic regression analysis showed that work shifts and workload simultaneously had significant influence on fatigue. In conclusion, shift and workload affect the severity of fatique experienced by co-assistants at Prof. Dr. R. D. Kandou General Hospital, Manado. Night shifts and heavy workload are predictors of increased fatigue.
