Antibiotic resistance has driven the search for natural antibacterial agents to treat acne vulgaris caused by Propionibacterium acnes. This study aimed to evaluate the anti-acne efficacy and secondary metabolite composition of sea cucumber (Holothuria nobilis) extract from Malalayang Dua Waters, Manado. Extraction was performed via the maceration method using 95% ethanol. Active compounds were identified through phytochemical screening, while antibacterial activity was tested using the paper disc agar diffusion method at concentrations of 50%, 75%, and 100%. Phytochemical screening revealed that the extract contained alkaloids, flavonoids, saponins, and tannins. The antibacterial assay yielded inhibition zones of 10.4 mm (50%), 11.5 mm (75%), and 12.2 mm (100%), respectively, all falling into the strong category. In conclusion, the ethanol extract of Holothuria nobilis exhibits strong potential as a natural antibacterial agent against Propionibacterium acnes through the synergistic mechanisms of its secondary metabolites.
