AKTIVITAS ANTIBAKTERI DARI TERIPANG LAUT YANG DI PEROLEH DI PERAIRAN BUNAKEN

Abstract


Sea cucumber or sea cucumber is one of the marine fauna that lives in shallow sea areas that are included in members of thorn-skinned animals (Echinoderms). Sea cucumbers produce secondary metabolites which are usually used as self-defense. Sea cucumbers are capable of producing bioactive compounds that are beneficial to humans, namely antibacterial. The purpose of this study was to test the antibacterial activity of the crude extracts of the three sea cucumbers, Holothuria (Halodeima) atra, Bohadschia marmorata, Pearsonothuria graeffei crude extracts. and partition fraction of sea cucumber Pearsonothuria graeffei against Staphylococcus aureus and Escherichia coli bacteria. Extraction begins with maceration of the sample using 95% methanol for three times and then the filtrate obtained is evaporated using a rotary vacuum evaporator. The extract obtained was fractionated by the Liquid Partition method using n-hexane, ethyl acetate, and methanol as solvents. The antibacterial activity testing method used was agar diffusion (Kirby and Bauer disc diffusion) with a concentration of 100 mg/ml of the test material and the amount of test material in a 30 l disc. The positive control used was Amoxsan for S. aureus bacteria and Ceftriaxone for E. coli bacteria while the negative control used methanol. For each bacterial medium the test was carried out with three replications and then the results were averaged. From this study, it was found that the three extracts of the sea cucumber samples and the three fractions of the sea cucumber P. graeffei were able to inhibit the growth of E. coli and S. aureus bacteria with moderate strength. The crude extract of B. marmorata showed the best activity against E. coli bacteria with an inhibition zone of 10 mm. Furthermore, crude extract of H. (H.) atra showed the best activity against S. aureus bacteria with an inhibition zone of 9 mm.
