ABSTRACT
Dengue virus (DENV) is the causal agent of dengue fever that is transmitted by Aedes aegypti and Aedes albopictus mosquitoes. DENV has four serotypes that have been widely recognized in the world: DENV-1, DENV-2, DENV-3 and DENV-4. DENV has RNA as the genome, and its length is almost 11 kilo bases. DENV RNA composed of 5'- and 3'-UTR; 3 structural genes; 7 non-structural genes (NS). Among the seven non-structural genes exist, NS3 plays an important role in viral replication as protease, helicase and NTPase. This study aimed to compare the variation of NS3 gene and amino acid sequences of DENV-2 in Indonesia with DENV-2 from several other countries. This research is an in silico study by using softwares to analyze the data retrieved from GenBank. The result of the multiple alignment of 100 sequences showed that NS3 gene from Indonesia samples had variations at 10 amino acid sites, which were occured at sites number 14, 119, 122, 171, 248, 289, 308, 392, 453 and 535, while the remaining global samples had 69 amino acid sites that were found to be variated.

ABSTRAK
Virus Dengue (DENV) merupakan penyebab penyakit demam berdarah yang ditransmisikan melalui nyamuk Aedes aegypti dan Aedes albopictus. DENV memiliki empat serotipe yang telah dikenal secara luas oleh dunia, yaitu DENV-1, DENV-2, DENV-3 dan DENV-4. DENV merupakan virus dengan materi genetik berupa rantai RNA yang panjangnya hampir 11 kilo basa. RNA DENV tersusun atas 5’- dan 3’-UTR; 3 gen struktural; 7 gen non-struktural (NS). Di antara ketujuh gen non-struktural yang ada, NS3 merupakan gen yang berperan penting dalam replikasi virus, yaitu sebagai protease, helikase dan NTPase. Penelitian ini bertujuan untuk membandingkan variasi sekuens dan asam amino gen NS3 DENV-2 yang ada di Indonesia dengan DENV-2 dari beberapa negara lain. Penelitian ini merupakan studi in silico dengan menggunakan software yang menganalisis data yang diperoleh dari GenBank. Hasil penjajaran 100 sekuens menunjukkan bahwa gen NS3 sampel Indonesia memiliki variasi yang terjadi pada 10 situs asam amino, yaitu pada nomor 14, 119, 122, 171, 248, 289, 308, 392, 453 dan 535, sedangkan sampel global memiliki 69 situs asam amino yang mengalami variasi.
