Abstract: Enterococcus faecalis exhibits a high prevalence in root canal treatment (RCT) failure. Although 2,5% NaOCl is effective in eliminating Enterococcus faecalis, it can cause apical tissue irritation and has an unpleasant odor. Herbal ingredients offer antibacterial properties with lower toxicity levels. Frangipani leaves (Plumeria rubra L.) contain antibacterial compounds such as flavonoids, phenols, and alkaloids. This study aimed to determine the effectiveness of frangipani leaf extract (Plumeria rubra L.) in inhibiting the growth of Enterococcus faecalis. This was a true experimental in vitro laboratory design using the well diffusion method. Frangipani leaf extract was obtained through a 96% ethanol maceration process. Treatment groups consisted of various concentrations of frangipani leaf extract (20%, 40%, 60%, 80%), a positive control (2.5% NaOCl) and a negative control (aquadest). Enterococcus faecalis cultures were obtained from the FMIPA Laboratory at Universitas Sam Ratulangi. The results indicated based on the mean diameter of the inhibition zones, the inhibitory strength from the highest to the lowest was: the positive control, 80%, 60%, 40%, 20% of frangipani leaf extract, and negative control. The Kruskal–Wallis test showed a p-value <0,05 indicating significant differences in the mean inhibition zone diameters across all groups. In conclusion, Frangipani leaf extract (Plumeria rubra L.) is effective in inhibiting the growth of Enterococcus faecalis with the 80% of extract being the most effective and classified as having strong inhibitory activity.
