ABSTRACT
Catharanthus roseus (L) G. Don also called Tapak dara plant is cultivated plant, besides its function as ornamental plant but also can be functioned as medicinal plant. Alkaloid compounds from this plant has been isolated and used as an anti cancer drug, are vinblastine and vincristine. Vinblastin and vincristine were poor alkaloid compounds produced in plants. Vincristine made from joined catharantine and vindolin, cause it is expected that vindolin be able to increase the content of vincristine. The research was carried out to knew the vincristine content of callus culture were given tryptophan treatment and vindolin. The research plan used was completely random design, by tryptophan combination (0, 75 and 150 mg/L) and Vindolin (0, 0,2, 0,4 and 0,6 mg/L), factorial. The vincristine content was determined  by high performance liquid chromatography (HPLC). Based on HPLC data, it was made standar curve and regression linear equation that describe the relationship between standar vincristine concentration of area. The obtained result from the research proved the tryptophan and vindoline treatment affecting the vincristine content. The highest vincristine content was showed to T1V0 treatment (Tryptophan 150 mg/L and Vindolin 0 mg/L), is 2,352 µg/g dw.

ABSTRAK
Catharanthus roseus (L) G. Don yang disebut juga tanaman tapak dara merupakan tanaman yang banyak dibudidayakan selain sebagai tanaman hias juga berkhasiat obat. Senyawa kelompok alkaloid dari tanaman ini telah diisolasi dan dijadikan obat anti kanker, yaitu vinblastin dan vinkristin. Vinblastin dan vinkristin adalah senyawa alkaloid yang sangat kecil dihasilkan dalam tanaman. Produksi alkaloid dari Catharanthus roseus (L) G. Don  Vinkristin terbentuk dari gabungan katarantin dengan vindolin, karena itu diharapkan dengan vindolin dapat meningkatkan kandungan vinkristin. Penelitian dilakukan untuk mengetahui Kandungan Vinkristin pada Kultur Kalus Catharanthus roseus (L) G. Don yang diberi Perlakuan Triptofan dan Vindolin. Rancangan penelitian yang digunakan adalah Rancangan Acak Lengkap (RAL) secara faktorial dengan perlakuaan kombinasi triptofan (0, 0,75 dan 150 mg/L) dan vindolin (0, 0,2, 0,4 dan 0,6 mg/L). Kandungan Vinkristin ditentukan dengan menggunakan metoda Kromatografi Cair Kinerja Tingggi (KCKT). Berdasarkan hasil KCKT dibuat kurva standar     dan persamaan regresi yang menggambarkan hubungan antara konsentrasi vinkristin standar terhadap luas area. Hasil yang diperoleh dari penelitian ini membuktikan perlakuan triptofan dan vindolin mempengaruhi kandungan vinkristin. Kandungan vinkristin tertinggi berada pada perlakuan T1V0 (Triptofan 75 mg/L dan Vindolin 0 mg/L), yaitu sebesar 2,352 µg/g bk.
