ABSTRACT
           Cancer is a health problem since it is difficult to cure and causes death for the patient. In cancer occurs uncontrolled cell proliferation and loss of ability to activate apoptosis program so that cancer cells become immortal. The purpose of this study was to obtain data on standardization of specific parameters and anticancer activity test against colon cancer cells (WiDr) from ethanol extract of seagrass Enhalus acoroides. Seagrass that has been taken from the habitat was then extracted by maceration method using 96% ethanol for 24 hours then repeated 3 times for 24 hours. Standardization of specific parameter data includes plant identity, organoleptic test, soluble compound in certain solvents, chemical content test and determination of flavanoid level. The anticancer activity test was using MTT assay method and continued with live cell percentage calculation then probit analyze and linear regression. Result of calculation of probit analysis got IC50 value from ethanol extract of seagrass against WiDr cancer cell equal to 2969.0 µg/mL and IC50 value of doxorubicin as positive control was equal to 9.215 µg/mL. The result of linear regression calculation obtained that IC50 value of ethanol extract of seagrass was 2067 µg/mL and IC50 doxorubicin value is 121.54 µg/mL. Result of this study concluded that according to the National Cancer Institute, the ethanol extract with series of concentrations ranging from 500 µg/mL did not have anticancer activity against colon cancer cells (WiDr), compared to the positive control doxorubicin.

ABSTRAK
Kanker merupakan masalah dibidang kesehatan karena sukar disembuhkan dan menyebabkan kematian bagi penderita. Pada kanker terjadi proliferasi sel yang tidak terkendali dan hilangnya kemampuan mengaktifkan program apoptosis sehingga sel kanker menjadi immortal. Tujuan penelitian ini yaitu mendapatkan data standardisasi parameter spesifik dan uji aktivitas antikanker terhadap sel kanker kolon (WiDr) dari ekstrak etanol Lamun (Enhalus acoroides). Lamun yang telah diambil dari habitatnya kemudian diekstraksi dengan metode maserasi menggunakan pelarut etanol 96% selama 24 jam kemudian di remaserasi sebanyak 3 kali selama 24 jam. Data standardisasi parameter spesifik mencakup identitas tanaman, uji organoleptik, senyawa terlarut dalam pelarut tertentu, uji kandungan kimia dan penetapan kadar flavonoid. Uji aktivitas antikanker menggunakan metode MTT assay dan dilanjutkan dengan perhitungan persentase sel hidup kemudian dilakukan analisis probit dan regresi linear. Hasil perhitungan analisis probit didapatkan nilai IC50 dari ekstrak etanol Lamun terhadap sel WiDr sebesar 2969,0 µg/mL dan nilai IC50 doxorubisin sebagai kontrol positif sebesar 9,215 µg/mL. Hasil perhitungan regresi linear diperoleh nilai IC50 ekstrak etanol Lamun sebesar 2067,53 µg/mL dan nilai IC50 doxorubisin sebesar 121,54 µg/mL. Hasil penelitian ini menyimpulkan bahwa menurut Nasional Cancer Institute ekstrak etanol dengan seri konsentrasi dimulai dari 500 µg/mL tidak memiliki aktivitas antikanker terhadap sel kanker kolon (WiDr), dibandingkan dengan kontrol positif doxorubisin.
