ABSTRACT
This study aims to optimize and validate the analytical methods in determining the total phenolic content of green gedi leaf extract (Abelmoschus manihot L.). Optimization is done by varying the volume of Folin-Ciocalteu reagents is 1.5; 2; and 2.5 mL with some concentration variation is 25; 50; and 75 % and Sodium Carbonate (Na2CO3) with variation of solvent volume 1.2; 2.0; and 4.0 mL with some variation of concentration 5.0; 7.5; and 10 %. Validation parameter of analysis method is done by determining accuracy, precision, LOD and LOQ, and linearity. The optimization result of analysis method showed that 1.5 mL of Folin-Ciocalteu reagent 25 % and 1.2 mL of Sodium Carbonate (Na2CO3) 5 % showed maximum absorbance of 0.432. Validity values of validation parameters of analytical methods obtained accuracy results at concentrations of 30, 40, 50 µg/mL, respectively is 88.786; 93.635; 83.636 %, precise with the value of Relative Standard Deviation (RSD) equal to 0.06 %, LOD equal to 22.363 µg/mL and LOQ 74.545 µg/mL and linearity obtained correlation coefficient r2= 0.993. The total content of phenolic in the green gedi leaf extract of Manado, North Minahasa, and Southeast Minahasa respectively are 913.65 ± 0.005; 788.16 ± 0.009; and 286.35 ± 0.001 µg GAE/g. While the total phenolic content in the fraction of n-hexane, ethyl acetate and aquadest were Manado 129.09 ± 0.017; 440.9 ± 0.009; and 55.45 ± 0.003 µg GAE/g, North Minahasa 88.18 ± 0.002; 260 ± 0.002; and 38.18 ± 0.003 µg GAE/g and Southeast Minahasa 82.72 ± 0.014; 168.18 ± 0.037; and 20 ± 0.004 µg GAE/g. As concluded that the concentration, volume and solvent used produce the optimal total phenolic content and validity values that meet the requirements to give reliable results.

ABSTRAK
Penelitian ini bertujuan untuk mengoptimasi dan memvalidasi metode analisis dalam penetapan kandungan total fenolik ekstrak daun gedi hijau (Abelmoschus Manihot L). Optimasi dilakukan dengan memvariasikan volume reagen Folin-Ciocalteu yaitu 1,5; 2; dan 2,5 mL dengan beberapa variasi konsentrasi yaitu 25; 50; dan 75 % dan Natrium Karbonat (Na2CO3) dengan variasi volume pelarut 1,2; 2; dan 4,0 mL dengan beberapa variasi konsentrasi 5; 7,5; dan 10 %. Parameter validasi metode analisis dilakukan dengan menentukan nilai akurasi, presisi, LOD dan LOQ, serta linearitas. Hasil optimasi metode analisis menunjukkan bahwa 1,5 mL reagen Folin-Ciocalteu 25 % dan 1,2 mL Natrium Karbonat (Na2CO3) 5 % menunjukkan absorbansi yang maksimal yaitu sebesar 0,432. Nilai validitas dari parameter-parameter validasi metode analisis memperoleh hasil akurasi pada konsentrasi 30, 40, 50 µg/mL secara berturut-turut yaitu 88,786; 93,635; 83,636 %, presisi dengan nilai Relatif Standar Deviasi (RSD) sebesar 0,06 %, LOD sebesar 22,363 µg/mL dan LOQ 74,545 µg/mL serta linearitas diperoleh koefisien korelasi r2= 0,993. Kandungan total fenolik pada ekstrak daun gedi hijau kota Manado, Minahasa Utara dan Minahasa Tenggara secara berturut-turut yaitu 913,65 ± 0,005; 788,16 ± 0,009; dan 286,35 ± 0,001 µg GAE/g. Kandungan total fenolik pada fraksi n-heksan, etil asetat dan aquades secara berturut-turut yaitu Manado 129,09 ± 0,017 ; 440,9 ± 0,009; dan 55,45 ± 0,003 µg GAE/g, Minahasa Utara 88,18 ± 0,002; 260 ± 0,002; dan 38,18 ± 0,003 µg GAE/g serta Minahasa Tenggara 82,72 ± 0,014; 168,18 ± 0,037; dan 20 ± 0,004 µg GAE/g. Konsentrasi, volume serta pelarut yang digunakan menghasilkan kandungan total fenolik yang optimal dan nilai validitas yang memenuhi persyaratan.
