ABSTRACT
Gedi (Abelmoscus manihot L.) is a tropical plant of the malvaceae family, has traditionally been long known in North Sulawesi as a vegetable. The purpose of this research is to know the optimization of the analysis method with several parameters tested that is concentration and volume of reactant and time of incubation, to know the validation of the analytical method used so as to give the right result in determining the total flavonoid content, and to determine the total flavonoid content of green gedi flavonoid (Abelmoscus manihot L.). Quantitative analysis method of flavonoids by using UV-Vis Spectrophotometer was used in this study. Optimum result was obtained that the optimum result was (AlCl3) 2% 0,5 mL reacted with acetic acid (CH3COOH) 5% 5 mL with the most optimal incubation time was 20 min. The validation parameters are precision, accuracy, linearity, LOD (Limit of Detection) and LOQ (Limit Of Quantitation) using UV-Vis Spectrophotometer at a maximum wavelength of 415 nm. The result obtained from the precision test fulfilled the requirement with RSD/KV equal to 0,00313%, with accuracy value which also fulfill the value (80-110%) that is 106,766%, the result of the correlation coefficient (r) is 0,993 and LOD value of 249,3 ppm and LOQ value obtained for 831 ppm. The total content of flavonoids in Manado City was 155,83 mg QE/g extract ± 0,0007, North Minahasa sub-district of 675,85 mg QE/g extract ± 0,049 and Tomohon City of 450,86 mg QE/g extract ± 0,032.

ABSTRAK
Gedi (Abelmoschus manihot (L.)) merupakan tumbuhan tropis famili malvaceae, secara tradisional telah lama dikenal di Sulawesi Utara sebagai tanaman sayuran. Penelitian ini bertujuan untuk mengetahui optimasi metode analisis dengan beberapa parameter yang diujikan yaitu konsentrasi dan volume reaktan serta waktu inkubasi sampel yang digunakan, dan mengetahui validasi metode analisis yang digunakan sehingga memberikan hasil yang tepat dalam penentuan kandungan total flavonoid, serta menentukan kandungan total flavonoid daun gedi hijau (Abelmoscus manihot L.). Metode analisis kuantitatif flavonoid dengan menggunakan Spektrofotometer UV-Vis dipakai dalam penelitian ini. Hasil optimasi metode analisis diperoleh bahwa hasil yang paling optimal adalah (AlCl3) 2% 0,5 mL direaksikan dengan asam asetat (CH3COOH) 5% 5 mL dengan waktu inkubasi yang paling optimal adalah 20 menit. Parameter validasi metode adalah presisi, akurasi, linearitas, LOD dan LOQ menggunakan Spektrofotometer UV-Vis pada panjang gelombang maksimum 415 nm. Hasil yang diperoleh dari uji presisi memenuhi syarat ketentuan dengan nilai RSD/KV sebesar 0,00313%, dengan nilai akurasi yang juga memenuhi syarat nilai (80%-110%) yaitu 106,766%, hasil dari koefisien korelasi (r) adalah 0,993 dan nilai LOD sebesar 249,3 ppm serta nilai LOQ yang diperoleh sebesar 831 ppm. Hasil kandungan total flavonoid pada kota Manado sebesar 155,83 mg QE/g ekstrak ± 0,0007, kecamatan Minahasa Utara sebesar 675,85 mg QE/g ekstrak ± 0,049 dan kota Tomohon sebesar 450,86 mg QE/g ekstrak ± 0,032.
