ISOLASI DAN UJI ANTIBAKTERI JAMUR ENDOFIT DARI TUMBUHAN MANGROVE Soneratia alba DI PESISIR KOTA MANADO

ABSTRACT
Symbiosis of endophytic fungi mutualism with plant tissue is able to produce secondary metabolite compounds similar to their host. The study aimed to isolate, characterize and test the antibacterial effectiveness of endophytic fungi isolates isolated from S. alba mangroves. Methods used include planting explants in isolation and testing antibacterial effectiveness with the modified Kirby-Bauer method. The results of the study obtained five endophytic fungi isolates isolated from mangrove S. alba with characteristics of fungi isolated macroscopic both color, shape, texture and speed of growth and microscopically with the discovery of hyphae that is blocked, branched and transparent and some isolates with spreading spores. Antibacterial testing showed all five endophytic fungi isolates positively inhibited the test bacteria. The highest Sone 3 isolate inhibited E. coli growth by 26.67 mm and the Sone 9 isolate provided the highest resistance to S. aures bacteria by 32.33 mm and against P. aeruginosa by 32.33 mm. This suggests all five fungal isolates have antibacterial compounds.

Simbiosis mutualisme jamur endofit dengan jaringan tumbuhan mampu menghasilkan senyawa metabolit sekunder yang mirip dengan inangnya. Penelitian ini bertujuan untuk mengisolasi, mengkarakterisasi dan menguji efektivitas antibakteri isolat jamur endofit yang diisolasi dari mangrove S. alba. Metode yang digunakan meliputi tanam eksplan dalam isolasi dan uji efektivitas antibakteri dengan metode Kirby-Bauer yang dimodifikasi. Hasil penelitian diperoleh lima isolat jamur endofit yang diisolasi dari mangrove S. alba dengan karakteristik isolat jamur yang berbeda secara makroskopis baik warna, bentuk, tekstur serta kecepatan pertumbuhannya dan secara mikroskopis dengan ditemukannya hifa yang bersekat, bercabang dan transparan serta beberapa isolat dengan spora yang menyebar. Pengujian antibakteri menunjukkan kelima isolat jamur endofit positif menghambat bakteri uji. Isolat Sone 3 paling tinggi menghambat pertumbuhan E.coli sebesar 26,67 mm dan isolat Sone 9 memberikan hambatan tertinggi terhadap bakteri S.aures sebesar 32,33 mm dan terhadap P. aeruginosa sebesar 32,33 mm. Hal ini menunjukkan kelima isolat jamur memiliki senyawa antibakteri.
