This study aims to determinate the the total phenolic, total carotenoids and antioxidant activity of extract of Manado Yellow corn seeds. The study initiated by extracting the powder of corn seed by using Soxhletation with several of solvents (hexane, ethyl acetate, buthanol, and methanol). Then continued with the determination of the total content of phenolics and carotenoids, DPPH radical scavenging activity and antioxidant capacity by using UV-Vis spectrophotometry method. The results showed that the ethyl acetate extract had the highest total phenolic content followed by butanol, methanol and n-hexane. The total phenolic content was 149.28; 59.90; 48.16 and 44.08; mg/kg, respectively. The results also showed that ethyl acetate extract has the highest carotenoid content, free radical scavenging activity and total antioxidant capacity than butanol, methanol and n-hexane extracts. These results suggest that ethyl acetate extract is very rich in phytochemicals and antioxidant properties.

ABSTRAK
Penelitian ini bertujuan untuk menentukan total fenolik, total karotenoid dan aktivitas antioksidan dari ekstrak biji jagung Manado Kuning. Penelitian dimulai dengan mengekstraksi serbuk biji jagung menggunakan metode sokhletasi dengan beberapa pelarut (heksana, etil asetat, butanol, dan methanol). Kemudian ditentukan kandungan total fenolik, karotenoid, aktivitas penangkalan radikal bebas DPPH dan kapasitas total antioksidan dengan metode spektrofotometri UV-Vis. Hasilnya menunjukkan bahwa ekstrak etil asetat memiliki kandungan total fenolik tertinggi diikuti ekstrak butanol, methanol dan n-heksana yaitu 149,28; 59,90; 48,16 dan 44,08; mg/kg. Hasil penelitian ini juga menunjukkan bahwa ekstrak etil asetat memiliki kandungan total karotenoid, aktivitas penangkalan radikal bebas dan kapasitas total antioksidan tertinggi daripada ekstrak heksana, etil asetat, butanol dan metanol. Hasil penelitian ini menyarankan bahwa ekstrak etil asetat sangat kaya dalam fitokimia dan bersifat antioksidan.
