ISOLASI DAN KARAKTERISASI BAKTERI POTENSIAL PENDEGRADASI OLI MESIN BEKAS DARI BEBERAPA LOKASI BENGKEL DI KOTA MANADO

ABSTRACT
Soil pollution due to indiscriminate disposal of used oil in workshops is currently getting more serious, this is due to the compound content in the oil which is difficult to decompose and can cause disease. This study aims to isolate, characterize and identify potential bacteria that degrade used engine oil from several workshop locations in Manado City. This research was conducted for 4 months, December 2020 to March 2021. Selection of bacteria using selective media (Busnhell-Haas) modified with the addition of used oil. The results showed that there were 3 isolates that were successfully isolated, namely bacteria 23, 29 and bacteria 33 with different characteristics and had been identified and it was found that bacteria 23 was a species of bacteria Citrobacter freundii which has the potential as bacteria to degrade used engine oil for the bioremediation process.

Pencemaran tanah akibat dari pembuangan oli bekas secara sembarangan pada bengkel-bengkel saat ini semakin serius, hal ini dikarenakan kandungan senyawa yang ada dalam oli yang sulit untuk diurai dan dapat menyebabkan penyakit. Penelitian ini bertujuan untuk mengisolasi, mengkarakterisasi serta mengidentifikasi bakteri potensial pendegradasi oli mesin bekas dari beberapa lokasi bengkel di Kota Manado. Penelitian ini dilaksanakan selama 4 bulan, Desember 2020 sampai Maret 2021. Seleksi bakteri menggunakan media selektif (Busnhell-Haas) yang dimodifikasi dengan penambahan oli bekas. Hasil penelitian menunjukkan bahwa terdapat 3 isolat yang berhasil diisolasi, yaitu bakteri 23, 29 dan bakteri 33 dengan karakteristik yang berbeda-beda serta telah teridentifikasi dan ditemukan bahwa bakteri 23 merupakan spesies bakteri Citrobacter freundii yang berpotensi sebagai bakteri pendegradasi oli mesin bekas untuk proses bioremediasi.
