ABSTRACTMiana leaf (Coleus scutellarioides [L] Benth.) contained flavonoids, tannins, triterpenoids,
steroids and essential oils are able to provide an antibacterial effect. The purpose of the study
was to made a balm extract from Miana leaf (Coleus scutellarioides [L] Benth.) and test the
effectivenen against scar infected bacteria in rabbit skin. Proces of balm extract from Miana
leaf has qualified the standard and tested on rabbit with 5 treatment groups, is injuries without
negative control, positive control, Miana leaf ointment 20%, 40% and 80%. All the rabbit cut
for 1,5 cm and given 0,2 mL of bacteria Staphylococcus aureus. The wounds were applied
with ointment three times daily. Observation was conducted everyday for 8 days. All data
was tested statistically using ANOVA (Analysis Of Variant) followed bay LSD (Least
Significant Difference) test. The result shows differences that the Miana leaf formulation
meets the ointment test requirement according to Farmakope Indonesia Edition III, wounds
were narrowed, scabs were formed and then wounds were closed. Statistical test showed that
there were significant effects on wound healing in rabbit, F count > F table (37,167 > 5,19).

ABSTRAK
Daun Miana (Coleus scutellarioides [L] Benth) memiliki kandungan flavonoid, tanin,
triterpenoid, steroid dan minyak atsiri yang mampu memberikan efek antibakteri. Penelitian
ini bertujuan untuk membuat salep dari ekstrak daun Miana (Coleus scutellarioides [L]
Benth.) dan uji efektivitas sediaan salep terhadap pengobatan luka yang terinfeksi
Staphylococcus aureus pada kulit kelinci. Pembuatan salep ekstrak daun Miana yang telah
memenuhi syarat dan diujikan pada hewan uji kelinci sebanyak 5 ekor dengan perlakuan,
dasar salep berlemak (kontrol negatif), Gentamicin salep (kontrol positif), salep daun Miana
20%, 40% dan 80%. Semua kelinci disayat sepanjang 1,5 cm dan diberikan bakteri
Staphylococcus aureus sebanyak 0,2 mL. Luka diolesi tiga kali sehari dengan salep yang
diuji. Pengamatan luka dilakukan setiap hari selama 8 hari. Semua data diuji secara statistik
menggunakan ANOVA (Analysis Of Variant) dan dilanjutkan dengan uji LSD (Least
Significant Different). Hasil penelitian menunjukkan formulasi salep ekstrak daun Miana
memenuhi persyaratan uji salep menurut Farmakope Indonesia edisi III. Luka sayat yang
dioleskan salep ekstrak daun Miana mengalami penyempitan luka, membentuk keropeng dan
menutup luka mulai hari ke-3 (tiga). Hasil uji statistik menunjukkan efek signifikan terhadap
penyembuhan luka pada kelinci, yaitu F hitung > F tabel (37,167 > 5,19).
