ABSTRACT
Sesewanua (Clerodendron squamatum Vahl.) empirically has been used by people in several areas in North Sulawesi to treat fever, fractures ,and reducing swelling. Sesewanua leaves contains flavonoids and alkaloids that can be potentially as an antioxidant. The purpose of this study was to determine the total phenolic content and antioxidant activity of the ethanol, ethyl acetate and n-hexane extracts from the leaves of sesewanua. The extraction of sesewanua leaves is using the maceration method. Determination of total phenolic contents use the Folin Ciocalteau method and testing of antioxidant activity using the 1,1-diphenyl-2-picrylhydrazyl (DPPH) method. Based on the results obtained, the highest total phenolic content ethyl acetate extract of 36.25 mg/L, followed by ethanol extract of 14.659 mg/L, and the n-hexane extract of 5.795 mg/L. Based on the calculation of the IC50, ethyl acetate extract has the highest antioxidant activity (13.084 mg/L), followed by ethanol extract (17.85 mg/L), and n-hexane extracts (23.737 mg/L).

ABSTRAK
Sesewanua (Clerodendron squamatum Vahl.) secara empiris telah digunakan oleh masyarakat di beberapa daerah di Sulawesi Utara untuk mengobati demam, patah tulang, dan penurun bengkak. Daun sesewanua mengandung flavonoid dan alkaloid yang dapat berpotensi sebagai antioksidan. Tujuan penelitian ini adalah untuk menentukan kandungan total fenolik dan aktivitas antioksidan ekstrak etanol, etil asetat, dan n-heksan dari daun sesewanua. Ekstraksi daun sesewanua dilakukan dengan menggunakan metode maserasi. Penentuan kandungan total fenolik menggunakan metode Folin Ciocalteau dan pengujian aktivitas antioksidan menggunakan metode 1,1-difenil-2-pikrilhidrazil (DPPH). Berdasarkan hasil yang diperoleh ekstrak etil asetat memiliki kandungan total fenolik tertinggi sebesar 36,25 mg/L, diikuti ekstrak etanol sebesar 14,659 mg/L, dan ekstrak n-heksan sebesar 5,795 mg/L. Berdasarkan hasil perhitungan IC50 ekstrak etil asetat daun sesewanua memiliki aktivitas antioksidan tertinggi dengan nilai IC50 sebesar 13,084 mg/L, diikuti ekstrak etanol sebesar 17,85 mg/L, dan ekstrak n-heksan sebesar 23,737 mg/L.
