Spons Phyllospongia lamellosa mampu menghasilkan senyawa bioaktif yang berpotensi sebagai antibakteri. Penelitian ini bertujuan untuk mengukur aktivitas ekstrak dan fraksi spons Phyllospongia lamellosa terhadap pertumbuhan bakteri Gram positif Staphylococcus aureus dan Gram negatif Escherichia coli di perairan Pulau Manado Tua. Sampel di ekstraksi menggunakan metode maserasi dengan pelarut etanol 95% dan selanjutnya difraksinasi menggunakan pelarut yaitu n-heksan, kloroform dan methanol dengan metode fraksinasi cair-cair. Uji aktivitas menggunakan metode disk diffusion Kirby dan Bauer. Hasil penelitian menunjukkan bahwa hasil pengukuran rata - rata diameter zona hambat ekstrak dan fraksi spons Phyllospongia lamellosa terhadap bakteri Staphylococcus aureus yang paling besar dihasilkan oleh fraksi kloroform dengan nilai rata – rata 15,96 mm ± 0,98 dan Escherichia coli oleh fraksi kloroform dengan nilai rata – rata 17,48 mm ± 4,80 sehingga keduanya dikategorikan kuat.
